中文摘要: 铁皮石斛(铁皮枫斗)是我国名贵中药材,素有“救命仙草”之称,由于其价格昂贵、石斛碱成分含量低,现行质量评价指标成分为含量较多的多糖,因此普遍存在用其他品种石斛冒充铁皮石斛的现象,铁皮石斛真伪鉴别手段有待提高。本课题以表面增强拉曼散射(SERS)原位检测技术为手段,基于铁皮石斛中石斛碱的组织部位含量分布特征,结合磁分离提取、富集石斛碱有效成分,提出了基于SERS光谱扫描成像指纹整合有效成分原位分布特征信息的方法来研究铁皮石斛专属性鉴定及定位含量测定验证模式。通过SERS成像原位指纹图库的建立,获得铁皮石斛与伪品具有鉴别特征的原位分布信息;选择调控磁性Fe3O4@SiO2@Au@Ag纳米粒子为SERS活性基底,建立基于磁分离的定位含量检测方法,获取SERS基底调控石斛碱含量相关的效率信息。通过数据处理与整合研究,挖掘明确铁皮石斛的鉴别信息基础,建立原位、定位的铁皮石斛真伪鉴别方法。

英文摘要: Dendrobium officinale is precious traditional Chinese medicine, known as “life-saving immortality” said. As the quality evaluation component is polysaccharide, so the phenomenon of counterfeiting Dendrobium officinale with other species of Dendrobium is prevalent. Based on characteristics of dendrobine content distribution in tissue sites, we use SERS (surface-enhanced Raman scattering) imaging technology to study the authenticity of identification in situ and the positioned content determination. Through the establishment of in situ SERS imaging fingerprint graphic database, we can obtain the identifying characteristics of in situ distribution. We selectively regulate magnetic Fe3O4@SiO2@Au@Ag nanoparticles as the SERS active substrate, and then establish the method of location detection based on magnetic separation mechanism. Finally, this study recommended the “in situ identification” to domestic and foreign pharmaceutical industry in the form of SERS imaging and provide a new authenticity identifying method for other valuable Chinese herbal medicines.
