中文摘要: 甲型流感病毒感染诱导的细胞因子风暴是引起急性呼吸窘迫综合症和多器官衰竭的重要原因。主要穹窿蛋白(MVP)是穹窿蛋白复合物的一个组成部分,它在调节细胞耐药性、细胞应激反应、肿瘤发生等方面起到重要作用。目前尚无研究报道MVP与甲型流感病毒诱导的细胞因子风暴之间的关系。本项目在前期工作的基础上,拟开展以下两部分研究:第一部分是利用小鼠模型和细胞模型研究MVP在甲型流感病毒诱导的细胞因子风暴中的作用。第二部分是研究MVP与调控细胞因子风暴的转录因子之间的相互作用以及对应的调控机制。力图明确MVP对甲型流感病毒诱导的细胞因子风暴的影响,阐明该基因调控细胞因子风暴的机制,为抗流感病毒药物的研发提供新信息和策略。

英文摘要: Influenza A virus-induced cytokine storm is an important cause of acute respiratory distress syndrome and multi-organ failure. Major vault protein (MVP) is the major constituent of vaults and is involved in multidrug resistance, nucleocytoplasmic transport and tumorigenesis. However, little is known about the role of MVP in influenza A virus-induced cytokine storm. Based on our previous work, we want to study the following two parts: The first part is to find the role of MVP in influenza A virus-induced cytokine storm by using mouse model and cell model. The second part is to study the interaction between MVP and transcription factor invloved in cytokine storm, and the corresponding regulatory mechanism behind it. This project is to find out the influence of MVP in influenza A virus-induced cytokine storm and the mechanisms accounting for it, providing new information and strategies for the development of anti-influenza drug candidates.
