中文摘要: Tourist-like MITE和Stowaway-like MITE在毛竹基因组分布广泛,本课题组已有的实验证据表明这两类MITE可能广泛参与了宿主基因表达调控,因此本课题计划在原有的工作基础上,1)大规模克隆毛竹MITE,调查MITE在基因组分布特性,分析MITE在毛竹基因表达调控网络形成过程中的参与机制;2)分析MITE在毛竹生长发育和形态变异过程拷贝数和插入位点的变化,调查MITE附近基因的表达谱变化,阐明MITE对毛竹生长发育和形态变异的调控作用;3)分离活性MITE,调查MITE所携带的调控元件和编译的小RNA对下游基因和靶基因表达模式的影响;4)选取能显著影响基因表达模式的活性MITE,转化拟南芥和水稻,在拟南芥和水稻不同发育阶段和组织器官中,诱导MITE在基因组频繁转座和大量增殖,调查外源MITE对宿主基因表达网络的重塑作用,阐明MITE与宿主基因表达调控网络互作机制。

英文摘要: Tourist-like miniature inverted repeat transposable elements (Tourist-like MITE) and Stowaway-like MITEs are very wide-spread in Phyllostachys pubescens and are possibly common involved into the regulation of host gene expression. The project plans to, 1) large-scale characterize the Tourist-like MITEs and Stowaway-like MITEs in P. pubescens, investigate the distribution characteristics of both of the MITEs in P. pubescens genome, and reveal the participation mechanism of MITEs into the regulatory network of host gene expression; 2) investigate the dynamic change of the copy number and insertion sites of MITEs during the growth and development,morphological variation of P. pubescens, analyze the influence of MITEs on host gene expression pattern, reveal the regulation mechanism of MITEs on the growth and development,morphological variation of P. pubescens; 3) isolate the active MITEs, and analyze the impact of the regulatory sequences contained in MITEs and small RNA encoded by MITEs on the expression of the downstream genes and target genes; 4)determine ative MITEs that could significantly regulate gene expression pattern, transform the active MITEs into Arabidopsis and Oryza sativa, induce the active MITEs frequent transposition and proliferation in Arabidopsis and O. sativa genome during the growth and develo
