中文摘要: 小青龙汤治疗哮喘经典而有效,本课题组曾对小青龙汤加减方做相关分子生物学、病理学方面的动物实验研究,发现其能有效地减轻哮喘小鼠气道重塑及炎症因子的沉积。但参与哮喘发病的炎症因子甚多,而气道炎症启动因子胸腺基质淋巴细胞生成素(TSLP)在哮喘发病过程中起到关键作用,而TSLP的表达受到IL-1β和 TNF-α等促炎细胞因子影响,经NF-κB调控与细胞表面受体相结合从而发挥炎症效应,以激活DCs,激发CD4+T 细胞前体向Th2转化,诱发更多的TSLP产生,这一信号环路可能解释哮喘发病的重要机制。本课题拟通过小青龙汤对哮喘小鼠气道炎症启动因子TSLP信息传导环路(TNF-α-NF-κB-TSLP-Th2-TNF- α)的干预研究,并对靶向细胞DCs表面Th2表达的影响进行体外实验,以发现小青龙汤对哮喘的治疗靶点,进一步阐释小青龙汤温肺化饮治疗哮喘的作用机制。

英文摘要: It is an effective and classic method to treat asthma with Xiaoqinglongtang. We have finished animal experiments of Xiaoqinglongtang from the view point of molecular biology and pathology,and found that Xiaoqinglongtang could effectively alleviate airway remodeling and reduce the deposition of inflammation factors in asthmatic mice. Generally, a number of inflammation factors are involved in the development of asthma, and the initiation factor thymic stromal lymphopoietin (TSLP) plays an essential role. The expression of TSLP is influenced by proinflammatory cytokine, such as IL-1βand TNF-α. Then, through the regulation of NF-κB, it binds to a receptor on the surface of the cell to induce inflammation. Consequently, the DCs are activated and the CD4+T cell precursors change into Th2, resulting in the generation of more TSLP.This signal pathway could explain the main pathogenesis of asthma. So we plan to find out the impacts of Th2 in vitro experiments. Thus, we can find the therapeutic target of Xiaoqinglongtang for asthma. In addition, this research can be used to further explain the mechanism of Xiaoqinglongtang in warming lungs, reducing phlegm and treating asthma.
