中文摘要: 角质膜(层)是植物叶片、花冠等地上器官特有的一层由酯类物质等组成的薄膜,是植物抵御不良环境的第一道屏障,其结构和组成成分与生物学功能密切相关,特别是角质膜外蜡质,是当前植物抗性研究的热点之一。尽管早先的研究指出角质膜可以反射紫外光辐射,但对于叶片角质膜如何减弱UV-B辐射的机理尚不十分清楚,尤其在非模式草本植物中报道很少。本项目拟选择宁夏干旱半干旱区分布的乌拉尔甘草为研究对象,以本课题组探索建立的人工模拟UV-B辐射的方法为处理手段,分别采用酶法分离角质膜技术、核磁共振分析和多变量数据处理技术、定量荧光PCR技术等,研究不同UV-B辐射处理模式下,叶片角质膜形态结构特征、代谢物图谱特征及其变化、角质膜蜡质及角质合成等相关的基因表达等,探索从不同层次认识甘草叶片对UV-B辐射增强的响应机制及可能的调控途径,为阐明干旱半干旱生境的植物进化及对环境适应过程奠定理论基础。

英文摘要: The cuticle is a translucent film covering the aerial parts of plants such as leaves and corollas, consisted of hydrophilic and hydrophobic polymeric, and act as the first protective barrier against the varied forms environment stress. Its biological function is closely connected with itself structure and components, especially the outmost waxex layer,which has been focused on by researchers. Although previous researches showed that cuticule could reflect ultraviolet rays, the mechanism about cuticule attenuation of UV radition has been unclear, especially in non-mode plants. The aim of the research project is to investigate the function of cuticle to attenuate short-wave UV radiation and mechanism of adaptation to UV-B radiation for Glycrryhiza uralensis, by applied NMR to analyze metabolites profiling, qRT-PCR analyze related genes, SEM and TEM to measure cuticle structure as well as waxes crystal. With this research we will able to characterise the responses mechanism to UV-B radiation for the leaves and cuticles of G.uralensis. Furthermore, the results will provide a theoretical basis for plant evolution and adaptation in arid and semi arid environment.
