中文摘要: 吸烟成瘾与烟碱在体内的代谢过程密切相关。因此,怎样实现烟碱代谢的活体动态监测并同步分析烟碱引起的神经药理学效应,成为研究的重中之重。本项目拟以正常Wistar大鼠、烟碱依赖-戒断综合症大鼠和烟碱代谢酶抑制剂处理大鼠为研究对象,建立烟碱及其代谢物活体分析的微透析-UPLC-MSn-稳定同位素示踪方法,对烟碱在大鼠脑内及外周血中的分布与代谢特征进行动态分析与比较,结合代谢关键酶CYP450活性的检测,深入探讨烟碱在动物模型体内的代谢规律;同时采用微透析-HPLC-ECD同步分析烟碱代谢对大鼠脑内重要神经递质的释放与代谢变化,获得有效评价烟碱神经药理学效应的指标;在此基础上,综合分析不同个体烟碱代谢的差异及其与重要神经递质变化的关系。为全面了解烟碱的代谢规律以及进一步阐释烟碱依赖的形成机理提供基础性数据支持。

英文摘要: Smoking addiction is closely related to the metabolic processes of nicotine in human body, and the study of the association between nicotine metabolism and its synchronized neuropharmacological effects becomes more and more important. This project intends to study the distribution and metabolic characteristics of nicotine in the brain and peripheral blood of free-moving Wistar rats, nicotine dependence-withdrawal syndrome rats and metabolic inhibitor-treated rats, by using microdialysis-UPLC-MSn method with the stable isotopetracer technique, which will be followed by the determination of CYP450 activity in the liver and brain; At the same time, the microdialysis-HPLC-ECD method will be applied to analyze the release and metabolic changes of neurotransmitters in the animal brains, to evaluate the neuropharmacological effects of nicotine metabolism. With comprehensive analysis of the differences in individuals nicotine metabolism and its relationship with neurotransmitter changes, this study will provide basic data to explain the mechanism of nicotine metabolism and nicotine dependence.
