中文摘要: 淋巴转移是胰腺癌转移到主要方式,也影响预后的重要因素,虽然目前的放化疗对其有一定的作用,但疗效不尽人意,更缺乏术前同时诊断和治疗的有效方法。随着纳米诊治技术的诞生,针对胰腺癌淋巴转移,同时诊断和治疗的难题可望得到解决。在既往研究基础上,我们设计合成Fe3O4修饰的磁性石墨烯纳米颗粒(GO-Fe3O4),利用其特殊的理化特性,研究其在磁共振成像(MRI)和染料-双模态下胰腺癌区域性淋巴转移的示踪能力;以胰腺癌淋巴道转移动物模型和胰腺癌原位淋巴转移动物模型为研究对象,以磁性修饰的纳米GO-Fe3O4为载体,开展其在胰腺癌淋巴转移中示踪和光热治疗的实验研究,进而验证磁性纳米GO-Fe3O4的光热治疗可行性、有效性和生物安全性。GO-Fe3O4纳米淋巴诊治系统集双模态示踪和光热治疗为一体,兼行胰腺癌淋巴转移的诊断和治疗,国内外目前还未见报道,为胰腺癌淋巴转移诊治提供新的策略和方法。

英文摘要: Lymph node metastasis is not only the main metastatic mode of pancreatic cancer, but also one of the most important prognostic factors. Although chemradiotherapy can improve the survival, the outcome of the pancreatic cancer patients remain poor. So far, we still lack accurate diagnosis method and an effective treatment method for lymph node metastasis in pancreatic cancer. With the application of theranostic nano-technology, the problem of diagnosis and treatment of the lymph node metastasis at the same time is expected to be solved. Based on our previous studies about the characteristics of lymph node metastasis of pancreatic cancer, we synthesized a functional nano-graphene (GO-Fe3O4) modified with Fe3O4. Using its special physical and chemical characteristics, to study the ability of magnetic resonance imaging (MRI) and dye - dual-modality mapping in the world regional lymph node metastasis of pancreatic cancer; With the delivery system of magnetic nano-graphene, the study of mapping regional lymph node and photothermal therapy was conducted to verify the excellent photothermal therapy efficiency and bio- safety on pancreatic cancer mice model with lymph node metastasis in the foot pad and orthotopic pancreatic cancer lymph node metastasis model . To our best knowledge, this is the first demonstration of using nano-graphene-based theranostic agent to image and treat pancreatic cancer lymph node metastasis. This study will provide us experimental evidence for a novel strategy and a new therapy for dual-modality mapping and photothermal ablation of regional lymph nodes in the orthotopic pancreatic cancer lymph node metastasis model with GO-Fe3O4 as a new nano-theranostic agent.
