中文摘要: 病毒病是影响我国油菜生产的重要病害之一。种植抗病品种是防治油菜病毒病的根本措施,但目前仍缺乏对病毒病高抗或免疫的油菜品种。在前期工作基础中,我们克隆了一个特异性应对病害防御反应信号的油菜基因BnSGS3,初步研究发现:病毒可诱导该基因上调表达;已获得BnSGS3基因RNAi载体转基因油菜,TuMV、CMV和TMV接种鉴定表明,转基因油菜体内BnSGS3基因表达受到抑制,植株对病毒更为敏感。为进一步明确该基因的功能及评估其抗病毒的效果,本研究拟构建BnSGS3的植物过表达载体,通过农杆菌介导的方法转化油菜和拟南芥SGS3基因缺失突变体,并对转基因植株进行病毒接种鉴定,通过分析BnSGS3的表达量和拷贝数与病毒抗性的关系,综合分析油菜BnSGS3基因的功能;研究结果为油菜抗病性遗传改良提供基因资源和理论指导,也将为其它类似植物-病原系统的研究提供重要参考。

英文摘要: Virus disease is one of the important diseases in oilseed rape (Brassica napus). Application of resistance varieties is an economical and effective strategy for control of the disease. However, varieties with high resistance or immunity to viruses are at present lack. Based on the preliminary work, we cloned a BnSGS3 gene from B. napus which specific induced by disease defensive reaction signal molecules. Pilot studies showed that viruses could induce BnSGS3 expression up-regulated. Transgenic oilseed rape plants with RNA interference vector of BnSGS3 had been obtained. They were more susceptible than non-transgenic controls inoculated by turnip mosaic virus, cucumber mosaic virus and tobacco mosaic virus when the expression of BnSGS3 was suppressed. In order to elucidate the function of BnSGS3 and estimate its resistance to viruses, in this study, plant over-expression vector of BnSGS3 will be constructed and transformed into B. napus L. and Arabidopsis thaliana SGS3 deletion mutant by using the Agrobacterium-mediated transformation. Transgenic plants will be inoculated with viruses. The role of BnSGS3 gene will be elucidated by analyzing the effect of the expression level and copy number of BnSGS3 on the resistance to viruses.
