中文摘要: 本项目的目标是以前期研究中筛选到的抑菌活性N-酰化苯并咪唑酮衍生物为先导化合物,通过分子优化设计和构效关系研究,获得抑菌活性优异、可供商化开发的抑菌活性化合物,初步阐明其抑菌作用机制。项目的内容包括:借助计算机辅助药物设计,利用虚拟组合化合物库、药效团匹配、3D-QSAR药效团模型进行分子优化,设计、合成,并系统评价这些化合物的离体和活体抑菌活性;通过测定线粒体呼吸链相关酶系的抑制作用,阐明其具体作用部位。项目的意义是:提出的化合物分子骨架新,结构修饰的余地大,可以开创出具有自主产权的新类型杀菌剂;N-酰化苯并咪唑酮衍生物虽然作用于真菌的呼吸系统,但其结构与现有作用于呼吸系统的杀菌剂(如甲氧基丙烯酸酯类,羧酰替苯胺类等)有明显差异,其作用部位可能不同于这些商品化杀菌剂,因此研究其作用机制在杀菌剂毒理学方面也具有一定的积极意义。

英文摘要: The objective of this investigation is to screen candidates based on N-acyl benzimidazolone derivatives for the development of agro-fungicide and clarify its mechanism. The contents of this study including: design, synthesis of benzimidazolone derivatives using virtual combinatorial libraries, pharmacophore matching and 3D-QSAR pharmacophore model and evaluating their antifungal activities. In addition, the preliminary mechanism of the compounds were studied by determination the activities against the composite enzyme of the mitochondrial respiratory chain. The project is characterized by the fact: N-acyl benzimidazolone deriatives have novel molecular skeleton and largely modification which can create a new type fungicide with independent property rights. Benzimidazolones as inhibitors of antifungal respiratory system but its structure have obvious difference with respiratory system Fungicides (such as Strobilurin and carboxylic acid anilide derivatives). It is suggesting that the active site of the benzimidazolones may be different from those of commercial fungicide. Thus, the study of its mechanism has certain positive significance in the fungicide toxicology.
