中文摘要: 癌症是世界上威胁人类健康的第一大疾病,早期诊断和治疗是目前癌症的治愈率最高的途径。肿瘤与正常组织相比具有血管壁间隙较宽、结构完整性差,淋巴回流缺失,微观环境呈弱酸性等特点,因此,目标设计一类pH响应高分子,其响应值通过调节聚合物疏水结构具有适应生物体微环境的任意响应值。首先,通过均聚物研究其pH响应值的影响因素,在均聚物研究基础上,进一步合成含PEO嵌段聚合物,得到一类碱性条件下可自组装形成胶束,在肿瘤和内涵体,溶酶体的弱酸性环境下解离形成单分子pH微调可控响应的纳米胶束。基于肿瘤组织的异型性,纳米胶束可具有选则性地滞留在肿瘤组织内部,在肿瘤的微观弱酸性环境下解离,通过包裹抗癌药物,在高分子侧基引入荧光探针,实现在肿瘤靶向性诊断的同时就释放药物。从而达到诊断治疗一体化的目的。

英文摘要: Diagnosis and therapy through the precise spatiotemporal control of agent delivery is an affective way for cancer therapy.Compared with nomal tissue, tumor tissue is reported to be a slight acidic enviroment, leaky vascellum and lack of lymphatic backflow. We plan to develop a set of reversible pH-sensitive polymers,their responsive pH value is tunable to be suitable for biological enviroment. Through the design and synthesis of a series homopolymers, the pH responsive value effects can be researched. Based on the homopolymer research, diblock copolymers containing hydrophilic PEO segment will be developed. These pH-activatable polymers with ionizable block segment will self-assemble as nanoparticles above their pKa and dissociate at acidic pH value because of the tunable hydrophobic groups. The tunable pH-responsive nanoparticles can accumulate at tumor tissue according to EPR effect and dissossiate to single molecules. Fluorescence probe will be conjugated to polymer backbone, hydrophobic drugs will be encapsulated into hydrophobic core. Cell detection and drug release can be achieved simultaneously.
