中文摘要: IL-18作为重要的细胞因子在衔接天然免疫和获得性免疫方面起重要的桥梁作用,参与多种自身免疫性疾病进展过程。IL-23和IL-17是新近发现的在多发性硬化(MS)中发挥重要作用的分子。初步研究发现,与对照组相比,MS病人外周血单个核细胞中IL-18、IL-23、IL-17表达明显上调,两者之间存在相关性,提示其在MS发病过程中可能发挥重要作用。关于IL-18及相关分子与IL-23、IL-17之间的调控网络在MS及其他自身免疫性疾病中作用迄今未见报道。研究组在上海建成MS监测网络,按照国际诊断标准进行MS确诊,已顺利完成上海123名患者的MS诊断,将建立样本库,运用分子流行病学方法,从分子、细胞和整体水平研究IL-18相关分子与IL-23、IL-17之间调控网络在自身免疫性疾病中的作用拓展我们对该网络在机体生理和病理过程中调节作用的认识,为更好地认识与治疗疾病提供理论和实验依据。

英文摘要: IL-18 is an important cytokine that plays a critical role in bridging innate immunity and acquired immunity, thus being found involved in the pathogenesis of many autoimmune diseases. IL-23 and IL-17 are recently discovered important cytokines in multiple sclerosis. In our preliminary studies, we found that compared with controls, the expression of IL-18 and IL-23/17 in peripheral lymphocytes of MS patients were significantly increased and highly correlated, implying that they play important roles in MS. There are no previous reports about the regulatory network between IL-18 and IL-23/17. We have established a database of MS patient in Shanghai, inclding 123 MS patients dignosed according to the international MS diagnose criteria (McDonald criteria). We are going to set up a databank with biological samples from MS patients, by using molecular epidemiological analysis, to investigate the effect of IL-18 and IL-23/17 network in autoimmune diseases. The results of this study will increase our knowledge about the network in autoimmune physiology and pathology, and provide theoretic and laborotary evidences to treat such diseases.
