中文摘要: 山药(Dioscorea opposita)是菜药兼用植物,其珠芽是重要的繁殖材料,但休眠期很长,限制了农业生产中适时播种。现有研究表明珠芽对生长调节剂的响应特殊,赤霉素延长休眠期,而多效唑等生长延缓剂则打破休眠,关于打破休眠的机理未见报道。本课题将研究多效唑的主要成分对休眠期山药珠芽的赤霉素受体基因GID1a、GID1b、GID1c,以及玉米素合成基因IPT1、IPT3、IPT5、IPT7表达的影响;研究上述基因的表达量、激素含量和山药珠芽休眠期长短间的关系;克隆休眠期差异大的山药和黄独(D. bulbifera)两份材料的赤霉素受体基因和玉米素合成基因的全长序列,分析对比cDNA序列特性,并预测蛋白质结构与相关性质。目的是弄清多效唑对休眠的调控是否与对这些基因表达的调控有关,探索这些基因与激素含量的关系,初步揭示多效唑打破山药珠芽休眠的机理,获得休眠相关的标记。

英文摘要: The bulbils of yam (Dioscorea opposita), an important plant used as vegetable and Chinese medicine, are important propagation materials, but the long dormant period of bulbils hinders planting yam in appropriate time. The bulbils respond to growth regulators distinguishingly, that is their dormancy is extended by gibberellin and is broken by growth retardants such as paclobutrazol. The mechanism of dormancy-break of the yam bulbil by paclobutrazol is still unknown. So we will study the effect of paclobutrazol on expression of the gibberellin receptor gene GID1a, GID1b,GID1c and the zeatin synthesis genes IPT1, IPT3, IPT5 and IPT7. Then the relationship between gene expression, hormone content and dormancy period will be studied. These gene sequences will be compared and the protein structure and properties will be divined after the genes of D. opposita and D. bulbifera being cloned. The purpose of this study is to find out whether paclobutrazol regulate these genes expression so it can regulate dormancy, to find the relationship between gene expression and hormone content, so as to preliminarily discover the mechanism of paclobutrazol breaking the dormancy, to abtain dormancy-related markers.
