中文摘要: 口腔成体干细胞研究在口腔再生医学研究领域具有广阔的前景。根尖牙乳头干细胞由于其具有免疫原性低,强大的群体倍增能力、增殖能力、端粒酶活性及成骨/牙本质潜能等优势而受到广泛的关注。研究发现CD24、碱性成纤维细胞因子等因子均与其stemness相关。本研究通过细胞培养,免疫组化,实时定量PCR及基因芯片分析等手段,分析根尖牙乳头干细胞在碱性成纤维细胞因子作用和CD24差异表达的情况下,Wnt/β-catenin信号通路调控SCAP干性的作用机制,并探讨Wnt/β-catenin信号通路与MAPKs通路在bFGF维持SCAP干性中的交通作用,旨在阐明维持或增强SCAP干性的机理,为将来组织工程中大规模利用牙源性干细胞进行扩大研究提供启示和思路,尤其是为SCAP应用在组织工程化牙髓牙本质及牙根的再生利用等转化再生医学研究提供新的理论基础和依据。

英文摘要: Oral adult stem cell research in the field of oral regenerative medicine research has a wide prospects.Stem cells from the apical papilla have low immunogenicity, strong proliferation ability, telomerase activity and osteogenesis/dentin potential advantages so get widely attention. Basic fibroblast growth factor and CD24 can maintain SCAP stemness, increase their differentiation potential.Studies shown that Wnt/beta - catenin signaling pathway has a close relationship between stem cell differentiation and self-renewal. This study use some methods, like cell culture, immunohistochemistry ,quantitative PCR , Western blot and gene chip techology to study whether the Wnt/beta-catenin signaling pathway play a important role in maintaining SCAP semness.We want to analyse the cross talk of Wnt/beta - catenin signaling pathway and MAPKs signaling pathway in enhancing SCAP stemness. We also want to clarify the role of Wnt/β-catenin signaling pathway in SCAP maintain its stemness of action. The reasearch can offere some useful datas and ideas to use oral dadult stem cell in tissue engineering.
