中文摘要: 血清或者血浆中小分子代谢物的含量测定是临床疾病诊断或健康水平评估的依据。NMR谱编辑技术无需对血清样品进行预处理就可以检测血清中代谢物的含量,是血清代谢物定量分析强有力的手段。但现有的NMR方法不能定量测定在血清中与蛋白质结合的代谢物。我们的初步研究表明,在血清中加入与血清蛋白质有强结合作用的小分子化合物,可使血清中某些代谢物信号大大增强。这就表明强结合小分子化合物可通过竞争结合原理置换出血清中与生物大分子结合的小分子代谢物,使结合态代谢物变成游离态代谢物,使其NMR信号由"不可见"变为"可见",从而使与血清蛋白有结合作用的代谢物的定量测定成为可能。本项目拟以与血清蛋白有强结合作用的小分子药物作为竞争结合配体,建立测定血清中与蛋白结合的小分子代谢物含量的NMR方法,这对于血清代谢物的定量检测、人类疾病诊断及健康水平评估等均具有重大的意义。

英文摘要: Quantitative determination of small molecule metabolites in blood serum or blood plasma is crucial to the clinical disease diagnosis or health assessment. NMR spectral-editing techniques are powerfull tools for quantitative analysis of serum metabolites because these techniques can quantify the metabolites in serum without the need of pretreatment. But existing NMR methods cannot quantify metabolites which bind to serum proteins. Our preliminary studies have shown that by adding small molecule compounds which bind strongly with serum proteins, some serum metabolites NMR signal is greatly enhanced. This indicates that metabolites which bind to biological macromolecules in serum can be replaced by strong binding small molecule compounds by competitive binding, and therfore turn bonded metabolites into free metabolites and NMR "invisible" signals into "visible". These make the quantitative determination of metabolites which bind to serum protein possible. Using small molecule drugs as competitive binding ligands, the project would build a NMR model for quantitative detection of bonded serum metabolites. It is of great significance to the human disease diagnosis and health assessment.
