中文摘要: 在临床肿瘤诊断中,联合多项肿瘤标志物指标同时检测,可实现对某特定癌症的准确诊断。因此,发展新型和简便灵敏的多种肿瘤标志物同时免疫测定具有重大的实际意义。本研究的目的是进一步拓宽同时检测分析物的数量,真正实现多指标检测,同时实现其快速、灵敏、便携的同时检测,建立多组分同时免疫测定的新方法。本研究在我们前期所建立的双组份同时免疫测定的基础上,提出把在单根电极上使用的多标记物模式结合到阵列电极中,使阵列电极的每根电极通过三明治的夹心免疫反应后捕获到不同的标记物,进而真正实现多指标检测;制备出具有更多分叉的电极,并做成电极阵列,发展无标记无试剂多组分同时快速免疫测定的新方法;设计新颖的信号放大策略,将带有不同电活性物质的标记物引入到电极表面测定,提高检测的灵敏度;将纳米阵列电极同微流控芯片相结合,发展低成本,便携的多组分同时检测,为多组分的同时检测提供新的途径。

英文摘要: In clinical analysis, simultaneous determination of multiple tumor markers associated with a particular type of cancer could provide a more reliable measurement. Thus, it is great important to develop new and simple formats for multiple markers analysis.The aim of this study is to further broaden the simultaneous detection amount of analytes, really achieving multianalytes detection. Besides, we want to develop the rapid, sensitive and portable simultaneous detection for multianalytes and establish a new method for simultaneous immunoassay.On basis of recent developed simultaneous immunosening for dual tumor markers by our, herein, we intend to introduce multiple labels into the array electrodes for achieving multianalytes detection; Also, we want to design the new array electrodes for high throughput, rapid and simultaneous multiple tumor markers analysis; At the same time, we plan to design the novel signal enhancement strategy for sensitive and simultaneous determination of multiple tumor markers; In addition, we try to combine array electrodes with microfluidic chip, development of low cost, portable and simultaneous multianalytes detection. This project will provide a new avenue for tumor marker simultaneous detection, and hold great potential for disease diagnostics.
