中文摘要: 油菜是我国最重要的油料作物之一,然而其种子油脂积累的分子机制及其调控网络仍不清楚。本项目在前期工作基础上,拟以油菜为主要研究对象,综合利用过量表达、RNAi、诱导表达、RNA原位杂交、酵母双杂交、BiFC、Co-IP和转基因等技术手段,探究BnTT8和BnTTG1转录因子在种子油脂积累方面的功能;通过解析它们对种子油脂积累相关农艺性状的影响、对油脂积累相关重要基因和转录因子的调控以及二者在此生物学过程中的互作关系,以期阐明它们对种子油脂积累的影响并初步判定他们的互作机制。本项目的开展不仅可以为揭示油菜油脂积累的分子机制及其调控网络提供重要的理论依据,而且可以为油菜等油料作物的高含油量和品质育种提供基因原件。

英文摘要: Oilseed rape (Brassica napus L.) is one of the most important oil crops in China. However, the molecular mechanisms and the regulatory networks regarding to seed oil accumulation are largely unknown.Based on our previous research work, many approaches inclusive of over-expression, RNAi, inducible expression system, RNA in situ hybridization, yeast-two hybridization, BiFC, Co-IP and transgenic techniques will be carried out to identify the functions of transcription factors BnTT8 and BnTTG1 in seed oil accumulation. This project aims to investigate the effect of BnTT8 and BnTTG1 on a series of agronomic traits related to seed oil accumulation, explore how they control the expression of the keys genes and transcription factors involved in seed oil accumulation and study their cross-talk in the regulation of this biological process. This will not only elucidate the molecular mechanisms and the regulatory networks of rapeseed oil accumulation, but also provide gene resources for increasing oil content and improving fatty acid compositions in oil crops,such as oilseed rape,via either traditional breeding or trangenic approaches.
