中文摘要: 盐度是十足目甲壳类幼体生物学研究的关键参数。三疣梭子蟹是重要的海洋甲壳动物。海水盐度变化尤其是盐度降低会引起应激并导致机体发生一系列变化。目前,有研究表明低盐胁迫会影响三疣梭子蟹的生长和存活,但关于低盐胁迫影响三疣梭子蟹免疫和生长的分子机制的研究尚未见报道。本项目拟采用转录组学、蛋白组学和代谢组学方法,分别在转录水平、蛋白翻译水平和代谢物差异等不同层次比较低盐胁迫与正常三疣梭子蟹的差异,以确定低盐胁迫影响三疣梭子蟹幼蟹生长和免疫的分子特征,阐明三疣梭子蟹适应低盐蟹胁迫的代谢调控网络,构建三疣梭子蟹低盐胁迫响应生物标志物体系。本项目利用后基因组时代最有效的高通量组学技术进行三疣梭子蟹低盐适应机制研究,将为海洋蟹类资源保护提供有力的科学依据。

英文摘要: Salinity is a key parameter for studying decapods. Portunus trituberculatus is an important marine crustacean. Change of the ambient salinity especially the decrease of salinity may induce shock and result a series of variation in marine animals. some reports demonstrated that low salinity can affect the growth and immune of the P. trituberculatus. However, report about molecular mechanism of low salinity affecting the growth and immune of P. trituberculatus was not found. In this program, methods including transcriptomics, proteomics and metabonomics will be designed to study the difference between normal and low salinity threatened P. trituberculatus. The purpose of this program is: 1) to elucidate the molecular characterization of low salinity stress affecting the growth and immune of juvenile P. trituberculatus; 2) to understanding of the regulation network of the gene expression and the molecular characterization of P. trituberculatus in response to low salinity stress; 3) to construct the system of biomarker for response of P. trituberculatus to low salinity. Application of the high-throughput technologies is an important step towards more detailed understanding the molecular mechanism of P. trituberculatus response to low salinity stress, which may provide potent theory basis for protection of marine crab resources.
