中文摘要: 生物节律对于新陈代谢等生物体正常生命活动的维持是必不可少的。CLOCK基因是哺乳动物中发现的第一个核心节律调节因子。以往对CLOCK研究多集中于其转录调控作用,而对其乙酰化酶活性的研究甚少。蛋白质组学研究表明,糖酵解/糖异生、三羧酸循环、氨基酸代谢、脂肪酸代谢途径的代谢酶乙酰化修饰呈现一定节律性。本项目拟探讨生物节律核心因子CLOCK是否以及如何通过乙酰化调节代谢通路及其生物学意义。

英文摘要: Circadian rhythms are essential for almost every aspect of normal physiology in human bodies, including metabolism. CLOCK is the first recognized mammalian circadian gene whose function was mainly considered as a transcription factor, but its effects as an acetyltransferase are not fully understood. Recent proteomic study found that acetylation of numerous metabolic enzymes in glycolysis/gluconeogenesis, TCA cycle, amino acid metabolism, and fatty acid metabolism exhibit rhythmicity. Hence, we will examine whether and how CLOCK acetylates metabolic enzymes to control the oscillation of cellular metabolism, and its biological implications.
