中文摘要: 阿胶是为我国药、食两用传统中药材,社会需求巨大。但近年来,由于毛驴的存栏数急剧下降,使得生产阿胶的唯一原料- - 驴皮产生了巨大的缺口;如何采用生物学手段提高驴皮的利用率和开发新的阿胶生产途径是急需解决的问题。驴真皮中的Ⅰ型胶原蛋白是阿胶的主要有效成分,其含量因驴皮的年龄不同而各异。 TGF-β1、CTGF基因是调控人体皮肤中胶原蛋白表达的关键基因,而对驴真皮中胶原蛋白的调控还未见报道。本项目将针对不同年龄段的新疆驴真皮组织,采用RT-PCR、免疫组化、Western blot技术,研究其Ⅰ型胶原蛋白的相对表达量与年龄、TGF-β1和CTGF基因之间的变化规律,为生产中选择最佳取皮制阿胶的驴龄提供依据;同时结合基因过表达及siRNA干扰技术,对TGF-β1、CTGF与胶原蛋白表达量之间的调控关系进行研究,为将来在体外建立驴胶原蛋白的高效生产体系,提供理论依据。

英文摘要: Ejiao,which is an important traditional chinese medicine for both medicine and foods,has enormous social need. Because the donkey population has declined sharply in recently years,donkey's hide,which is unique production material for Ejiao,has been far behind the requirement. It is urgent problem to resolve that improving utilization rate of donkey's hide and developing a new method to produce Ejiao. The typeⅠcollagen of doney dermis is the main effective constituent of Ejiao, which would be different in various age of donkey dermis. The TGF-β1 gene and CTGF gene are the key genes to regulate the expression of collagen in human skin. But for donkey, there is no relative report. This project will study changing regularity of type I collagen,age and the TGF-β1 and CTGF gene of Xinjiang donkey dermis using RT-PCR,Immunohistochemistry and Western blot. Our result will provided evidence to select optimal donkey's hide to produce Ejiao in production. At the same time,we will study the regulating relationship of the TGF-β1 and CTGF gene on collagen in donkey dermis using gene overexpressing method and gene siRNA technology. It will provided theoretical evidence to set up high efficient in vitro production system of donkey collagen.
