中文摘要: 以发现差异表达蛋白群为目标的蛋白质组学筛查分析需要高通量的分离分析方法。本研究拟建立高质量纳流色谱微柱的大规模、高通量、低成本制备技术,发展集高效富集-分离为一体的多功能柱型。在此基础之上,构建基于并行多道纳流液相色谱的高通量筛查分析系统,并联合液滴微流控技术实现超微量组分的大规模制备和目标组分提取。以临床样本(人尿液/血液)为对象,考察高通量筛查-目标蛋白群深度解析这一分析策略的有效性,进而为临床蛋白质组学研究提供有用的生物学信息。

英文摘要: Screening proteomics demands high throughput separation techniques for large-scale multiple sample comparison and discovery of differentially expressed protein panels. In this project, we will develop a high throughput, low cost and mass-production technology for preparation of high quality microcolumns for nanoflow liquid chromatography use. We will also develop multifunctional columns integrating high efficiency concentration and separation. Based on these developments, we will establish a multi-lane nanoLC platform for high throughput screening, in combination with a micropreparation and target sample isolation technology based on droplet microfluidics. Using real-world proteomic samples, e.g. human urine/serum, we will investigate the platform's effectiveness in high throughput screening and in-depth characterization of target proteins, in order to extract biologically significant information.
