中文摘要: 既往研究表明康复训练可以促进海马区神经干细胞的增殖和分化,但其具体分子机制尚不明了。Cx43半通道/缝隙连接是细胞与外界信息交流的重要通道之一,在脑缺血缺氧损伤中,起到重要的信息传递作用。我们前期研究表明康复训练可以促进缺血缺氧大脑Cx43和Wnt信号通路相关分子的表达,并且,近年来,在缺血缺氧心肌,人们还发现Cx43通过激活Wnt信号通路促进细胞再生,这提示我们,中枢神经系统是否也有相似机制存在?因此,本项目选用创伤性脑损伤模型(TBI),从离体和在体两个水平,研究3个问题:1.Cx43是否参与了TBI大鼠神经再生的调控?2.Cx43是否通过Wnt信号通路调控TBI后神经再生?3.康复训练是否通过Cx43促进大鼠TBI后神经再生。简而言之,即观察是否存在康复训练→Cx43→Wnt信号→神经再生信号通路。阐明以上问题,将为TBI疾病的康复治疗提供理论依据,具有重要的理论及临床意义。

英文摘要: Past studies showed that the rehabilitation training can enchance the neurogenesis greatly in the rats with hypoxia and ischemia damage. However,the molecular mechanism still remains unclear. Cx43 hemichannel/gap junction is one of the most important channels for communication between cells and extracelluar environments. In our early studies,we found the increased expression of Cx43 and Wnt signal pathway-related molecules.Moreover,recently,in the ischemic myocardium, researchers demonstrated Cx43 increased the cell regeneration via Wnt signal pathway.It suggested us the similar mechanism in the brain. Accordingly, in present study, we are inclined to elucidate three questions by traumatic brain injury model:1. 1.Is Cx43 hemichannel/gap junction involved in the regulation of neural regeneration after TBI? 2.Does Cx43 hemichannel/gap junction affect the neural regeneration through Wnt signal pathway ?3.Does rehabilitation training increase the neural regeneration through Cx43 hemichannel/gap junction? In short, we will detect if there is the signal route for rehabilitation→Cx43→Wnt signal pathway→neurogenesis. Our study will provide the theoretical basis for the rehabilitative treatment of TBI.
