中文摘要: 肿瘤疫苗的出现为恶性肿瘤的治疗开辟了新的思路。然而目前的疫苗体系由于肿瘤抗原免疫原性很弱,其治疗效果并不理想。本项目提出构建基于新型平面粒子氧化石墨烯(GO)的肿瘤疫苗输送体系,充分利用微米级GO的"平面折叠效应"及其在生物学/免疫学方面展现出的多种独特性质,发挥抗原递送和免疫激活的双重功能。该体系在不引入其它免疫刺激佐剂的前提下,简单而巧妙地融入了高效募集抗原提呈细胞、增加抗原摄取和促进抗原交叉提呈三个关键策略,实现"多点突破",程序性放大机体对肿瘤细胞的杀伤和识别能力,以达到彻底清除肿瘤的目的。该项目将为设计和研究基于新型纳微米粒子的高效、安全的肿瘤治疗方法提供科学依据。

英文摘要: Using vaccines delivery system shows promissing signs on tumor therapy. However, most vaccine antigens have poor immunogenicity, which largely compromise their effects. In our project, we propose to establish a tumor vaccine system based on the novel flat graphene oxide (GO). In terms of several unique properties in bio/immunological response, the "folding" micro-sized GO can perform a dual role of antigen carrier and immune stimulators. This vaccine system integrates three independent strategies in a quite facile manner to improve the antigen immunogenicity. Without addition of other immune adjuvants, the GO can also efficiently facilitate the recruitment of antigen presenting cells, improve the cellular uptake, and enhance the presentation of tumor antigens. Such a "One but All" system is expected to induce high level of anti-tumor responses in a programmable way and finally eliminate the tumors. This project will shed light on the incorporation of micro/nano particles in tumor therapy through a safe and efficient way.
