中文摘要: 脑卒中后抑郁(PSD)直接影响卒中患者的神经康复和治疗,降低生活质量,严重者促使患者死亡,但其发病的具体机制至今未能明确,临床抗抑郁药物治疗效果亦不尽如人意。根据最近提出的"抑郁症的神经可塑性假说",本研究试图以cAMP-PKA-CREB信号通路为切入点,建立PSD大鼠模型,实验纳入CREB基因剔除大鼠,通过观察PSD大鼠接受丰富康复训练(ERT)后,大鼠行为学及海马可塑性的变化,以及信号通路各因子的动态变化,由此证实ERT是否通过促进海马神经可塑性导致PSD大鼠抑郁的改善,并进一步探讨ERT改善PSD的分子机制及细胞内信号的转导途径,为预防和治疗PSD提供重要的实验室依据。

英文摘要: Post stroke depression (PSD) directly affected neurological rehabilitation and treatment of patients, reduced the quality of life, and even caused death of patients, but the specific mechanism has not been identified. The effect of clinical antidepressant drugs was not satisfied. According to the latest study of "neural plasticity hypothesis of depression", this study attempts to take cAMP-PKA-CREB signal pathway as the breakthrough point, and establishes a PSD rat model. The experiment includes CREB gene removed rats. The rats received enriched rehabilitation training (ERT). The testing index includes the change of rat ethology and hippocampal plasticity, and the expression change of each factor in signal pathway. It would confirm whether ERT promots the hippocampus plasticity and leads to improvement of depression on PSD rats through this results. To further discusses the molecular mechanism and cellular signal transduction of ERT on improving PSD, and provides important laboratory basis on the prevention and treatment of PSD.
