中文摘要: 实时检测技术应用于蛋白质组研究、药物筛选和疾病检测等领域中的小分子或弱相互作用研究时,一直受低信息量、低特异性和高检测成本的困扰。将相敏椭偏与压电传感相结合实现光-声联合生物分子实时检测,是本项目拟通过设计和实施的一种高信息量、高灵敏度、高特异性、多通道、快速检测的新方法。项目中通过构建倏逝波相敏特性椭偏,利用光波相位变化检测灵敏度比光强变化灵敏度高几个量级的优势,获得高检测灵敏度。同时结合压电传感技术可获得质量、粘度、密度、应力、粘弹性等样品参数,提高检测信息量和信号特异性。基于相敏椭偏与压电传感的光-声联合检测技术,研究生物分子相互作用动态检测方法,来实时跟踪检测分子组装、生物分子间结合、解离的整个动态过程。建立动力学模型对生物分子相互作用过程进行实时分析,以获得结合速率、解离速率和平衡常数及蛋白质分子亲合系数等动态参数。最后尝试从原ng/ml级降低至pg/ml的痕量检测。

英文摘要: Less Information and specificity and high cost of real-time analysis technology are not enough to study the interaction of small biomolecular or weak interaction in the field of proteome, drug screening and disease diagnosis. A high information, high sensitivity, high specificity, multi-sample and fast detection method is studied based on the combination of phase sensitive ellipsometry and piezoelectric technology in this project.Considered that ellipsometry is a phase sensitive technology, sensitivity will be extraordinarily improved by combining ellipsometry with evanescent wave. More information including mass, sticky, density, stress and viscoelasticity can be obtained by piezoelectric technology. Combined phase sensitive ellipsometry with piezoelectric technology, information and specificity of biomolecular interaction can be improved. Furthermore, the assemble, association and dissociation in the process of biomolecular interaction will be detected and analyzed in order to get the dynamic parameters including the association rate,dissociation rate and equilibrium constant based on the dynamic model of biomolecular interaction. Finally, the conditions corresponding to high sensitivity are obtained and the trace analysis from ng/ml to pg/ml is attempted.
