中文摘要: 水菖蒲是常用中药和民族药,民间流传用于糖尿病治疗,但未见文献记载。我们的前期研究发现,水菖蒲根乙酸乙酯提取部位ACE在体内外均有明显抗糖尿病作用,已获国家发明专利。ACE抗糖尿病作用机制多样,且对糖尿病动物胰岛具有保护作用。本申请项目基于肠道内分泌激素GLP-1在糖尿病治疗中发挥的重要作用,进一步阐明ACE对肠道内分泌和胰岛细胞的作用及分子机制。研究采用人工造模肥胖小鼠和糖尿病大鼠及体外培养肠道L细胞和胰岛β细胞,观察ACE的抗糖尿病和胰岛保护作用,探讨其与GLP-1分泌、表达和作用的相关性,并探索其调控信号通路。项目研究可为水菖蒲抗糖尿病作用及机制的全面阐明提供实验依据,为胰岛细胞保护机制的深入研究提供思路和方法。研究成果将为进一步的分子机制研究奠定基础,有利于提高我国中药抗糖尿病作用机制的研究水平,挖掘传统中药和民族药材的药用潜力,对中药现代化和新药发现具有重要的理论和实践指导意义。

英文摘要: Acorus calamus Linn, a commonly applied traditional Chinese drug and ethnic drug, is used in the therapy of diabetes in traditional folk medicine of America and Indonesia without any literature. We found the ethyl acetate fraction of Acorus calamus Linn (ACE) has obvious antidiabetic effects in vitro and in vivo, which has acquired the Chinese patent of invention recently. ACE has diverse mechanisms of antidiabetic effects and protects pancretic islets in the diabetic animal db/db mice. In this application, based on the importance of intesinal hormone glucagon-like peptide-1(GLP-1) in diabetes care, we will further elucidate the effects and molecular mechanisms of ACE in the intestinal endocrine cells and pancreatic islet cells. As to the experiment design, artificial experimental models of obese mice and diabetic rats will be employed in vivo studies, and intestinal endocrine L cells and pancreatic islet beta cells will be utilized in vitro studies. It will be observed the antidiabetic effects of ACE in different animals and the protection of the pancreatic islet cells, and also it will be discussed the relationship between the forementioned effects and the incretin GLP-1 and the signaling pathway regulating secretion, expression and function of GLP-1.
