中文摘要: 肺转移是骨肉瘤患者主要致死原因,关于其机制还不清楚。本实验组在前期研究中发现IGFBP5在高转移细胞系中低表达,过表达后抑制肿瘤生长和肺转移发生。随后我们过表达了IGFBP5不同的功能结构域,发现只有过表达C端结构域的肿瘤细胞侵袭能力下降。C端结构域是IGFBP5与细胞外基质(ECM)的结合区域,而ECM与肿瘤转移关系密切。因此我们推测IGFBP5是通过阻断了ECM与肿瘤细胞的相互作用而抑制肿瘤转移。为了进一步确定IGFBP5与骨肉瘤转移预后的相关性及阐明IGFBP5抑制骨肉瘤转移的机制,我们拟申请本课题首先利用免疫组化对骨肉瘤患者中IGFBP5的表达情况及与转移预后的相关性进行评估,在此基础上对IGFBP5通过抑制ECM-整合素 通路抑制转移的机制作初步探索。本研究将有助于利用IGFBP5进行转移风险度预测及理解骨肉瘤转移的分子机理,为研究新的治疗预防策略提供思路。

英文摘要: Osteosarcoma is the most primary malignent of bone and soft tissue with high metastasis rate to lung. To illustrate the mechanism of metastais of bone, we previously established cell sublines with different pulmonaty potential and identified IGFBP5 as the anti-metastasis gene by Microarray anylasis. futhermore, we overexpressed different domain of IGFBP5 in High-metastasis OSA cell lines and find only C-domain inhibit the invasion of tumor cells in vitro and in vivo. In current project, we plan firstly to collect the samples of osteosarcoma from Tangdu hospital and anylasis the expression of IGFBP5 in the tissue. Then, we will test the combination of IGFBP5 to ECM which block the Integrin pathway. we then culture the OSA cells with different ECM materials. meanwhile we use the antibody to block the Integrin pathway, invasion and adhesion test will be adminastrated to test the change of OSA cells in invasion ability in vitro. we finally transplant the IGFBP5 overexpressed cells with Integrin pathway blocked in nude mice. automatic metastasi will be observed by sacrifice the mice in 6 weeks.
