中文摘要: 大黄鱼是我国最具特色的海水养殖品种之一,溶藻弧菌是养殖大黄鱼最主要的致病菌之一,粘附作用是病原菌感染的第一个关键步骤。本项目在前期研究基础上,采用2种转座子构建溶藻弧菌的突变体库,通过粘附活性检测筛选粘附缺失突变菌株,进而克隆和鉴定粘附基因;通过综合比较野生型菌株、粘附突变菌株和遗传补偿菌株在生长速率、运动能力、对粘液的趋化能力以及粘附动力学等方面的差异,通过基因的表达产物对粘附作用的影响等方面研究来分析粘附基因在粘附作用中的具体功能;通过研究理化因子对粘附基因表达量的影响和对粘附活性的影响来探讨溶藻弧菌对大黄鱼肠粘液的粘附作用的基因调控。本项目的研究成果不仅有助于深化对溶藻弧菌的粘附机制和致病机制的认识,还可为大黄鱼养殖病害的抗粘附疗法提供重要的理论依据。

英文摘要: Large yellow croaker (Pseudosciaena crocea/Larimichthys crocea) is one of the main maricultured fish in China. Vibrio alginolyticus is one of the most important pathogens which has been commonly associated with the epidemic vibriosis and leads to mass mortality of cultured large yellow croaker. Bacterial adhesion to fish surfaces is an important step in the initial stage of infection. In this study, 2 transposons will be introduced to construct the mutant library of V. alginolyticus, and the adhesion defective mutants will be screened out by in vitro adhesion assay. The adhesion-related genes will be cloned and identified. In order to understand the role of each adhesion-related gene in the bacterial adhesion, the growth rate, motility, chemotaxis to muscus, as well as the adhesive kinetics of the wild type strain, mutants and the complemented strain will be detected and compared. To get a better understanding of the role of adhesion-related genes, the effect of the gene products on the bacterial adhesion will be detected. In order to probe into the genetic regulation of bacterial adhesion of V. alginolyticus to intestinal mucus of large yellow croaker, the environmental influence on the expression of adhesion-related genes and the bacterial adhesion will be investigated.
