中文摘要: 合成糖脂因具有分子结构及功能可调的特点引来越来越多的关注。糖脂是一种两亲性分子,作为细胞膜的成分,和磷脂、胆固醇等以双分子层形式构建细胞膜骨架,并在细胞表面分子识别、信息传输、膜融合等细胞反应过程中起到重要作用。糖脂是细胞表面标志性物质,也是细胞表面抗原的重要成分,因此,糖脂应用于药物、抗体或DNA载体,能够提高细胞识别力,增强靶向性。本项目拟利用糖脂的分子识别能力,以基因转染载体的设计、制备为应用背景,研究糖脂与阳离子磷脂混合脂质体的制备方法及其稳定性,研究糖脂分子结构和空间构象对脂质体及其双分子层膜性质的影响,探索糖脂/磷脂混合脂质体作为载体用于DNA运送、转染过程的机理,为设计和制备多功能型脂质体基因转染试剂以及脂质体的其它应用提供理论指导。

英文摘要: Synthesized glycolipids are attracted more and more attention due to their tunable molecular structure and function. Glycolipid is one of important biosurfactants. As a component of cellular membrane, it constructs the framework of cellular membrane in a bilayer state with phospholipid and cholesterol. It certainly acts important roles in many cellular reactions, such as molecular recognition, signal transfer, membrane fusion, etc. Meanwhile, glycolipid is also the symbolic substance and antibody component of cell surface. Thereby, introduction of glycolipid into carriers for drug, antibody and DNA is believed to enhance the targeting and increase the efficiency of transfection. The aim of this study is to design and prepare functionalized liposomes for gene transfection. Glycolipids with various chemical structures are added into LB membrane and liposomal bilayer of phospholipid. The effect of their chemical structure, and spatial conformation on properties of liposome and lipid bilayers is discussed. The mechanism of applying lipid/glycolipid liposomes to DNA delivery and transfection is explored. The study is hoped to provide theoretic directions for design and preparation of multifunctional gene carriers.
