中文摘要: 循环肿瘤细胞(CTC)检测在肿瘤的分期诊断、治疗方案的快速评估、肿瘤复发的监测以及药物开发等方面具有重大意义。如何从外周血中数亿个细胞中识别捕获少数的几个肿瘤细胞是当前CTC检测的瓶颈。本项目拟通过体外筛选技术,获得多条特异性识别上皮细胞黏附分子EpCAM及其它肿瘤细胞膜外标志物、肿瘤细胞的核酸适体,结合微流控高通量富集方法,实现外周血CTC的高灵敏、高选择性的快速富集检测。尽管EpCAM被广泛认为是CTC识别捕获的重要肿瘤标志物,基于EpCAM核酸适体的捕获富集方法仍未见文献报告。以核酸适体作为捕获探针,解决了抗体易失活、捕获效率低、成本高的问题。通过微流控芯片实现血液的高通量、全自动分析检测。项目的成功实施将为癌症扩散早期诊断和预警、治疗方案评估等提供重要的检测途径。申请人在核酸适体的筛选应用、微流控芯片的生物分析应用等相关领域具有扎实的研究基础,为项目的顺利实施提供了有力的技术保障。

英文摘要: Sensitive and selective methods for circulating tumor cell(CTC) detection are of great clinical significance in cancer prognosis, minimal residual disease detection, cancer therapy assessment, early diagnosis of invasive cancers and cancer drug development. Unfortunately, detecting a few CTCs among millions to billions of cells and differentiating the CTCs from epithelial non-tumor cells and leukocytes represent daunting technical challenges. In this project, aptamers against several cancer cell member biomarkers and cancer cells will be generated. A microfluidic chip functionalized with aptamers for CTC capturing,enumerating will be designed and fabricated for highly sensitive and selective CTC analysis from whole blood. The use of aptamers as affinity ligands to replace antibody avoids several problems associated with antibody methods such as protein denaturing, low capturing efficiency and high cost.On the other hand, microfluidics offers the advantages of high throughput and automated preconcentration&detection.There is no CTC detection method based on EpCAM aptamer reported so far, although EpCAM has been widely recognized as a key biomarker for CTC enrichment.
