中文摘要: 由意大利青霉侵染引起柑橘青霉病是柑橘采后重要病害,目前主要采用化学抑菌剂控制,存在很大安全隐患。研究新型、低毒、高效的柑橘采后抑菌剂对于柑橘的安全生产具有重要意义。本项目组前期研究发现5,7-二羟基黄酮类化合物是蜂胶抑制意大利青霉的活性成分,其中松属素的抑菌活性最强,对意大利青霉呼吸作用产生了直接的抑制作用。本课题拟在前期研究的基础上, 以松属素为先导化合物,分析松属素处理前后意大利青霉线粒体结构与功能、呼吸电子传递链、细胞凋亡等方面的生理生化变化,结合比较蛋白质组学技术检测松属素处理后菌体差异表达蛋白,确定松属素抑制意大利青霉的作用靶点和作用机理;进一步采用比较分子场分析法(CoMFA)研究5,7-二羟基黄酮类化合物与作用靶点之间的三维定量构效关系(3D-QSAR),构建3D-QSAR模型,为定向合成和设计新型、安全的柑橘采后抑菌剂提供实验参考和理论依据。

英文摘要: Citrus blue mold, caused by Penicillium italicum, is an economically important postharvest disease of citrus fruits. To prevent fruit decay, chemical antifungal agents such as sodium O-phenylphenate, thiabendazole, and imazalil, are routinely used either during preharvest or postharvest, which caused many food safety issues. Therefore, it is necessary to explore effective and eco-friendly, normally safe fungicide alternatives against these pathogens. In our previous reseach, we found 5,7-dihydroxyflavonoids and their analogues were the antifungal active compounds in propolis against P.italicum, and pinocembrin had the strongest antifungal activity and appeared direct inhibition on the respiration of P.italicum . On the basis of the previous investigations, the objectives of this project were to evaluate the physiological and biochemical changes of P.italicum after pinocembrin treatment in the structure and function of mitochondrion, respiratory electron transport chain, cell apoptosis and so on. Combined with the results of differentially expressed proteins by proteomics, the possible action target and action mode of pinocembrin on the P.italicum will be explored.
