中文摘要: 中国番茄黄化曲叶病毒(Tomato yellow leaf curl China virus,TYLCCNV)卫星DNA编码的βC1蛋白(TYLCCNB-βC1)是一个RNA沉默抑制子和症状决定因子,申请者已经确定与βC1蛋白互作的一个植物寄主因子为NtRFP。本项目拟围绕βC1蛋白与NtRFP的互作机制开展研究,鉴定NtRFP与βC1互作的结构域,确定NtRFP在病毒侵染和症状形成中的作用,研究NtRFP是否介导βC1蛋白的泛素化,并确定βC1蛋白的泛素化位点,明确βC1蛋白在普通烟中是否能被26S蛋白酶体降解,比较分析βC1蛋白不能被泛素化的突变体的致病性,从而明确植物通过NtRFP蛋白与βC1蛋白互作并介导其泛素化的生物学意义。上述工作的开展,可望了解双生病毒致病及寄主植物抗病毒机理。

英文摘要: The βC1 protein of Tomato yellow leaf curl China betasatellite (TYLCCNB) functions as a RNA silencing suppressor and pathogenicity determinant. Tobacco NtRFP is a host factor interacting with βC1 protein. The project will focus on the mechanism of the interaction between βC1 and NtRFP. We will dissect the domains necessary for βC1 and NtRFP interaction, effects of NtRFP on TYLCCNV/TYLCCNB infection and symptom appearance, whether NtRFP mediates βC1 ubiquitination and the ubiquitination sites of βC1, whether βC1 is degraded by 26S proteasomes in Nicotiana. tabacum cells, as well as effects of βC1 ubiquitination site mutations on virus infection and symptom appearance, so that we can assess the biological significance of βC1-NtRFP interaction and NtRFP mediating βC1 ubiquitination. The above results obtained in this study will uncover the mechanisms of the geminivirus pathogenicity and plant antiviral defense response.
