中文摘要: 胃肠平滑肌运动的主体是平滑肌细胞,外来神经、肠神经以及各种体液因素对平滑肌运动的调控,主要通过平滑肌细胞的离子通道或收缩蛋白来实现的。研究表明,炎症、氧化应激等可以诱导平滑肌细胞离子通道重建,引起离子通道电生理特性改变即电重建,最终导致收缩功能障碍。然而,目前在消化道动力障碍性疾病发病机理的研究中,有关平滑肌细胞电重建与动力障碍的关系,研究报道甚少。我们的前期工作表明,小鼠不全性肠梗阻使小肠平滑肌明显肥厚并伴有电生理特性和收缩功能异常,是理想的平滑肌细胞电重建模型。因此,本项目在小鼠不全性肠梗阻模型上,利用免疫组织化学、分子生物学以及电生理学等多学科技术,探讨平滑肌细胞电重建及其对平滑肌运动障碍的影响及分子机制,阐明平滑肌电重建是胃肠动力障碍的重要原因之一以及平滑肌细胞电重建的分子机制。本项研究具有重要的生理学与病理生理学的理论意义,也对诊断和治疗胃肠动力障碍性疾病具有潜在的临床意义。

英文摘要: Smooth muscle cells are the principal part in gastrointestinal motility and the extrinsic, enteric nerves and othter humoral factors can regulate smooth muscle motility via ion channels or contracting proteins in smooth muscle. Previous study indicated that inflammation, oxidative stress may induce ion channel remodeling in smooth muscle cell and change in electrophysiologic property which is namely electric remodeling and finally induced motility disorder. However, it is little reported about relationship between electric remodeling of smooth muscle cells and smooth muscle motility disorder in study of the mechanism in motility disorders of gastrointestinal smooth muscle. Our previous study demonstrated that intestinal obstruction induced smooth muscle hypertrophy and changes in electrophysiologic property and spontaneous contraction in partially small intestine obstruction mouse model, so it is a ideal model of smooth muscle cell electric remodeling. In present study, the effect of smooth muscle cell electric remodeling on smooth muscle motility and its molecular mechanism will be investigated by using immunohistochemistry, molecular biology and eledtrophysiology techniques and clarified smooth muscle cell electric remodeling is an important reason of motility disorders and remodeling mechanism in gastrointes
