中文摘要: 制备具有高稳定性、长循环特性、生物素化PEG修饰的包被SPIO的新型磁性脂质体(Biotinylated magnetoliposomes,Bt-MLs);测定其理化特性,体内分布及体内外抗巨噬细胞吞噬特性;评价其对荷肺癌裸鼠模型及微小肺癌灶的MRI靶向增强成像效果。本研究证实了Bt-ML具有超顺磁性、低毒性、抗网状内皮系统吞噬的特性,在体内实现了长循环,并初步证实了对裸鼠皮下种植型肺癌组织的T2负性被动及主动靶向成像作用;对利用磁共振早期定性肺部微小结节具有重要的作用,同时对其他肿瘤的受体磁共振靶向成像也具有重要的借鉴意义。

英文摘要: In the current study,we synthesized a novel kind of highly stable, long-circulating, biotinylated and PEG modified magnetoliposomes,with SPIO as core structure.Its physico-chemical properties,tissue distribution and anti-macrophage ability were evaluated.Finally,nude mice bearing lung cancer models were established to assess T2 negative and active enhancement effect of magnetoliposomes. In conclusion,Bt-MLs we synthesized reveal superparamagnetic and biocompatible features,low cytotoxicity , and good anti-macrophage ability in vitro and in vivo. Besides,they have long-circulating characteristic and T2 enhancement effect in the transplanted lung cancers by biotin-avidin signal amplification system.The novel MR agent may be useful for differentiating malignant from benign micro pulmonary lesions in early stage. Receptor-directed MR contrast agents will be potential to improve sensitivity and specificity of MR imaging and to detect other neoplasms early in the similar method.
