中文摘要: 黄檗为药材关黄柏的基源,国家二级保护植物。其生长缓慢,野生种群更新困难,遗传多样性正在逐渐丧失;长期以来关黄柏药材的短缺,在短时间内难以逆转。生长地不同的黄檗所产种子的品质和药材的质量存在差异,并且这种差异与生态因子有很强的相关性。本项目通过对黄檗分布区近100个位点生态因子的分析,2个位点的动态监测,进行生态特征定量评价,分析黄檗种子品质,特别是休眠特性形成与气象因子的关系;对药材中小檗碱、掌叶防己碱、药根碱、绿原酸、盐酸黄柏碱、木兰花碱、盐酸巴马汀、黄柏酮、黄柏内脂等8种成分进行计量,通过差异性分析建立药材质量与生态因子(气象和土壤)的相关性;研究种子在不同温度条件诱导下的基因表达谱,发现萌发途径关键酶基因并比较其表达差异。项目对揭示黄檗繁育的生态适应机制,预测野生种群幼苗的形成能力,推动优质药材的产业化生产,保护黄檗野生资源具有较大的理论及现实意义。

英文摘要: Phellodendron amurense Rupr., a second-class protected plant species in our country, is the basic source of the medicinal material Cortex Phellodendri amurensis. Howevr, it is hard to resolve the latter's urgent shortage for a long time because of the slow growth, increasingly loss of genetic diversity, and the difficulty of renewing wild populations of Phellodendron amurense Rupr. There is a significant difference on the quality of seeds and material of Phellodendron amurense Rupr from different habitats, which is related strongly to the ecology factors. In this grant, the correlation will be investigated by analyzing the characters of ecology factors for 100 habitats, dynamic monitoring two habitats and calculating the seed quality, especially the relationship between the weather and the formation of the dormancy of Phellodendron amurense Rupr. Also, eight ingredients of Phellodendron amurense Rupr, including berberine, palmatine, jatrorrhizine, chlorogenic acid, phellodendrine chloride, magnoflorine, Palmatine chloride, obacunone and obaculactone, will be calculated and the correlation between quality and ecology factors (weather and soil) will be set up. Besides, the expression profiling of genes induced by low temperature will be investigated in this grant and the genes coding important enzymes will be further discovered. The goal of the grant is to discover the adaptive mechanism for the Phellodendron amurense Rupr breeding and predict the viability of the wild seedlings. The finial goal of the grant is to promote the industrialization of excellent materials and protect wild sources of Phellodendron amurense Rupr.
