中文摘要: 草莓生产中花发育很重要。Agamous基因控制配子发生,对草莓育种有潜在的应用价值。本项目旨在利用转草莓FvAgamous基因第一个内含子(FvAGI)的植株这一人工创造的类似不育突变体的新种质,研究FvAgamous基因(FvAG)对草莓花发育的影响,明确内源FvAGI 非编码RNA(LncRNA)特性,从细胞学和分子生物学的角度,探讨转化FvAGI不育植株可能的机理。本项目是申请人前期研究的深入和延续,试材独特,内容新颖,研究成果可解析FvAG基因功能和内含子作用,丰富AG基因及非编码RNA调控花发育的生物学理论,并为理解植株不育提供科学依据;同时可促进利用AG基因创建草莓不育系的核心种质分子设计育种的发展,为作物不育分子设计育种提供新思路,具有重要的理论和实践意义。

英文摘要: The proposed project is aimed at a better understanding of the function of a long non-coding RNA and the mechanism of gene regulation mediated by the first intron of the AGAMOUS (AG) gene in strawberry Fragaria vesca. We identified a candidate long non-coding RNAs (LncRNAs) from the first intron of FvAG and showed that this LncRNA exhibits tissue- and stage-specific expression in strawberry flowers. The function of this LncRNA is not known and is suspected to play a role in regulating FvAG expression. We produced transgenic plants by over-expressing this first intron (FvAGI) under the control of the Cauliflower Mosaic Virus (CaMV) 35S promoter. The transgentic strawberry plants showed sterility. We hypothesize that RNA-directed DNA methylation, perhaps mediated by the LncRNA, is involved in the sterile phenotype of the transgenic plants. Further cellular and molecular biology experiments will be used to define the cause of transgenic plant fertility and the role of LncRNA of FvAG in fertility regulation.
