中文摘要: 近红外荧光探针的设计、合成及生物成像应用是当前化学和生物学等交叉学科的一个重要研究方向。核糖核酸(RNA)和过氧化氢(H2O2)都是与人类重大疾病相关的分子。针对目前所面临的缺乏追踪癌细胞内RNA附近H2O2的动态变化的近红外荧光探针的瓶颈问题,探针的研究需要构建高稳定性的近红外RNA/H2O2荧光探针,以满足当前生物医学研究的迫切需求。项目组具有RNA荧光平台及近红外荧光平台的研究基础,旨在发展良好稳定性的新型近红外RNA/H2O2荧光平台,并基于该平台构建能追踪癌细胞内RNA附近的H2O2的近红外荧光探针,并利用超分辨显微镜追踪癌细胞内RNA附近的H2O2的动态变化。本项目拟发展以咔唑为基本骨架的近红外荧光探针,该探针有望为解决目前近红外荧光探针所面临的无法追踪癌细胞内RNA附近H2O2的动态变化问题提供理论指导和实验依据, 具有重大的科学意义和应用价值。

英文摘要: Design, synthesis and biological application of near-infrared fluorescent probes for imaging are very important in the interdisciplinary field of chemisty and biology. RNA and hydrogen peroxide are associated with human major diseases. Currently there are some near-infrared hydrogen peroxide fluorescent probes, but the probes that can trace hydrogen peroxide near RNA in cancer cell are very little. So it is important to build near-infrared RNA/H2O2 fluorescent probe platform with high stability to meet the urgent need of current biomedical research. The project team has foundation in RNA fluorescent probe research and the probes have excellent performance in near-infrared imaging with high stability. We choose carbazole as the basic skeleton of the new near-infrared fluorescent probes. This project aims to develop novel near infrared fluorescent platforms that have good stability, and the probes based on the platform can trace hydrogen peroxide near RNA in cancer cell by super resolution microscope. Above all, this project will provide theoretical guidance and experimental basis for solving of the issue, and has important scientific significance and application value.
