中文摘要: 表观遗传在真核生物的发育过程中发挥重要的调控功能,其中DNA甲基化修饰主要由非编码小干扰RNA(siRNA)介导。siRNA的产生过程主要由RNA聚合酶IV参与,其定位过程则主要由RNA聚合酶V参与。RNA聚合酶IV/V的4/7亚基复合物最近被发现在siRNA介导的DNA甲基化途径(RdDM)中发挥重要的调控作用。本项目将重点研究RNA聚合酶IV/V调控表观遗传的分子机制,主要通过结构生物学方法解析Pol IV/V的4/7亚基复合物的晶体结构,揭示复合物与其同源蛋白在关键功能区域的变化,并利用生物化学方法寻找复合物特异性结合的核酸底物类型,深入研究复合物结合其特异性核酸底物的重要区域与分子机理,进而用生化方法阐明植物特异的RNA聚合酶IV/V及其4/7亚基复合物参与调控RdDM途径的结构基础和作用机制。本研究将加深对siRNA介导的DNA甲基化途径以及植物特异的RNA聚合酶的认识。

英文摘要: Epigenetics plays important regulatory functions during eukaryotic developmental process. One epigenetic mark, DNA methylation, is mostly directed by non-coding siRNA. RNA polymerase IV acts in the siRNA biogenesis process, while RNA polymerase V functions in the siRNA targeting process. 4/7 subcomplexes of RNA polymerase IV/V are found to be crucial for RdDM pathway regulation. Our project will focus on the mechanism that RNA polymerases IV/V regulate epigenetics, through determining the structures of 4/7 subcomplexes of RNA polymerase IV/V by crystallography and studying the differences in the crucial functional regions between plant 4/7 subcomplexes and their homologs. We will utilize biochemical methods to find out the specific nucleic acid substrates of 4/7 complexes, and clarify the important functional regions and mechanism of how the 4/7 subcomplexes bind the specific nucleic acid substrates, which aims to demonstrate the structural basis and mechanism that plant specific 4/7 subcomplexes regulate RdDM. Our project may help us understand siRNA-directed DNA methylation pathway as well as the plant-specific RNA polymerases.
