中文摘要: 分子医学的核心在于我们对细胞内外药物靶向体系的结构,相互作用,功能调控以及其与疾病关系的精确理解,已知至少有50%的药物靶标位于膜上。而膜蛋白的研究由于其两亲性,不均一性以及对膜上脂分子的依赖一直以来都是结构生物学的瓶颈。本项目立足于当今世界飞速发展的质谱分析技术,利用质谱方法固有的优势,结合非变性质谱,蛋白质组学方法,离子迁移质谱,以及脂分子定性定量技术等,寻求建立并完善多维一体化的生物质谱方法并将其应用于一系列目标膜蛋白复合物,以深入了解它们在细胞膜上的动态结构,功能调控,并探索脂分子对膜蛋白结构和功能的作用,力求多角度多层次描述生物分子复合物的相互作用机制,并由此探究它们在健康生物体正常功能和疾病相关功能故障的分子基础,为疾病诊断和药物的定向设计提供帮助与导向。

英文摘要: Central to molecular medicine are protein machineries which maintain homeostasis, and their roles in the cells of healthy and diseased individuals, of which 50% are membrane embedded. However many of these machineries continue to resist structural and dynamical elucidation due to their amphipathy, hetetrogeneity, and close partnership with membrane lipids for proper activity. Consequently the ultimate goal of a high resolution four dimensional view of the cell remains distant. Alternative strategy is therefore required at the frontiers of biomedical research. Here we propose an integrated mass spectrometry approach targeted at membrane protein complexes exploring every aspects ranging from their interactions with lipids to low resolution structural models. Our vision is to develop mass spectrometry and complementary techniques to investigate the structure, dynamics and interactions of membrane associated macromolecular assemblies in disease. As such we will strive to extend the approach from structural biology to molecular medicine.
