中文摘要: 本项目结合重金属磷光配合物长发射寿命的特点与共轭聚电解质的信号放大效应,拟设计、合成含铱配合物的磷光共轭聚电解质探针材料并实现其在生物传感、癌细胞靶向与光动力学治疗的应用。首先,我们将亲水性的基团引入到共轭聚合物的侧链来改善材料的水溶性,为其后续检测和成像奠定基础。通过改变聚合物中磷光铱配合物的含量,研究不同条件下聚合物主体到铱配合物客体的能量传递,为实现比率法的检测提供依据。其次,我们将特异性识别基团引入聚合单体中,实现高选择性的生物传感,利用磷光共轭聚电解质长发射寿命的优势,我们引入时间分辨技术扣除背景自发荧光信号的干扰,提高检测的信噪比和灵敏度。进一步将癌细胞靶向配体连接到共轭聚合物链上,实现共轭聚电解质对癌细胞的靶向标记。磷光共轭聚电解质作为光敏剂产生活性氧杀死癌细胞,实现光动力学治疗。

英文摘要: Considering the long emission lifetime of phosphorescent heavy-metal complexes (PHMCs) and the amplified signal output of conjugated polyelectrolytes (CPEs), it is quite interesting to graft PHMCs (as energy guest) into the CPEs (as energy host) to yield phosphorescent conjugated polyelectrolytes (PCPEs). In this proposal, we plan to design and synthesize a novel kind of sensing materials based on conjugated polyelectrolytes containing phosphorescent iridium (III) complexes, and to realize their applications in biosensing, bioimaging and photodynamic therapy. First, poly(ethylene glycol) (PEG) is grafted to the polymers to enhance their water-solubilities. Secondly, some special receptors of biomacromolecules, such as protein, are introduced into the monomers to achieve biosensing with high selectivity and competivity. Taking advantage of the long emission lifetime of PCPEs, time-resolved photoluminescent technique is applied in the sensing to eliminate the interferences from the background autofluorescence and enhance the signal-to-niose ratio and sensitivity, especially in the complicated media.
