中文摘要: 以DNA、RNA为核心的核酸系统,在分子层次起着处理信息的中心作用。生物分子电路是近年来计算与生物交叉产生的前沿研究领域。本项目拟利用DNA自组装、链置换和纳米颗粒等生物技术,研究复杂分子逻辑电路,构建纳米尺度的分子计算装置,探索分子接口电路实现对其他分子系统精确而程序化的检测与控制。重点研究内容包括:1)研究分子逻辑电路设计与分析的原理,包括核酸序列编码算法、分子信号重建机制以及分子电路与其他分子系统之间进行控制和数据信息的交换机制;2)研究稳定通用的分子逻辑门和触发器件,搭建较为复杂的分子逻辑电路,探索构建具有运算和存储能力的分子装置,设计生物实验验证方案,研究分子逻辑电路仿真方法;3)探索其在生化检测和医学诊疗等方面的应用,构建逻辑探针进行智能检测分析。这项研究将在纳米尺度控制、检测、加工以及程序疗法等方面有着潜在的应用,在计算、生物和医学之间构建一种新的联系。

英文摘要: Nucleic acid system with DNA, RNA as the core, plays a central role in information processing at the molecular level. In recent years,biological molecular circuit is a frontier research field at the cross area of computer science and molecular biology. This project intends to use the DNA self-assembly,strand displacement and nanoparticles to study the complex molecular logic circuit, eventually build molecular computational device at nano-scale, explore the construction of molecular interface circuit of other molecular system and procedures of the detection and control accurately. The key research contents include: 1) the principle of design and analysis of the molecular logic circuit, including nucleic acid sequences encoding algorithm, signal reconstruction mechanism and the exchange mechanism for control and data information between molecular circuit and other molecular systems; 2) study the stability of general molecular logic gates and flip-flops, construction of complex molecular logic circuit,design of biological experiment to test, exploring construction of molecular devices with computing and storage capacity finally, study the design of molecular logic circuit design and simulation modeling method; 3) explore the application in the biochemical detection and medical diagnosis etc.,construct a logic probe for intelligent detection and analysis. This study has potential application in nano-scale control, testing, processing and program therapy, and construct a new connection between the computation, biology and medicine.
