中文摘要: 高通量筛选系统是目前药物研发、生命科学和化学研究的重要工具。针对当前上述研究领域对高通量筛选系统微型化的迫切需求,在研究组前期发展的液滴实验室(DropLab)技术、微流控芯片加工和光学检测技术的基础上,研制具有自主知识产权的超微量、高通量、自动化微流控液滴分析筛选系统。将皮升级液滴定量量取和组装系统、高速三维移动平台系统、超高密度液滴阵列芯片、高灵敏荧光检测和数据处理系统,以及仪器控制、显示和电子电路系统均集成于该筛选系统中,实现整体系统的集成化和自动化,并将其应用于分子水平的药物筛选、蛋白结晶条件筛选和数字PCR等研究领域。另辟蹊径,打破国外公司在高通量筛选技术和仪器上的垄断,为我国科学仪器的战略性发展,也为我国在新药创制、结构生物学研究、组合化学合成、疾病早期诊断等领域的研究做出重要贡献。

英文摘要: High throughput screening systems are important tools for drug development and fundamental research of life science and chemistry. Toward the requirements for miniaturization of high throughput screening systems, the objective of the present project is to develop a ultramicro amount, high-throughput and automated droplet-based microfluidic analysis and screening system with completely independent intellectual property, on the basis of the DropLab, microfluidic chip fabrication and fluorescence detection techniques previously developed in the applicant’s group. Multiple modules including quantitatively sampling and assembling system for picoliter scale droplets, high-speed x-y-z translational stage, ultrahigh-density droplet array chip, high sensitive fluorescence detection and data processing system, as well as instrument control, display and electronic circuit systems will be integrated into the screening platform to realize the integration and automation of the system. The analysis and screening system will be applied in the fields of drug screening on molecular level, protein crystallization condition screening, and digital PCR.
