中文摘要: 近年来,全基因组关联研究(Genome-wide association study, GWAS)已发现十几个与结直肠癌发病风险密切相关的易感区段。然而,由于研究手段及成本的限制,目前对这些易感区段还未开展大规模的深入研究。为了挖掘这些区段内隐藏的更多功能性基因和位点,本研究拟采用全新的精细定位策略,首先对GWAS发现的10q22.3结直肠癌易感区段(以易感位点rs704017为中心)内全部基因进行功能分析,寻找与结直肠癌发病或进展相关的易感基因。再瞄准发现的易感基因调控区进行精细定位,利用病例-对照的关联研究,挖掘易感区段内潜藏的更多功能性结直肠癌易感位点。本研究以基因功能研究为基础,可以对GWAS发现的结直肠癌易感区段进行更为有效的精细定位,为发现全新的功能性遗传变异提供了新思路和新方法。所鉴别的遗传变异在未来可能为结直肠癌癌风险预测和高危人群筛检等个体化防治提供新的线索和依据。

英文摘要: Several susceptible loci have been identified through genome-wide association study (GWAS) of colorectal cancer. However, the functional basis underlying these SNPs/loci has not been well elucidated. In order to identify more functional genes or SNPs in these susceptible loci, in this proposal, we will perform a functional analysis of genes in 10q22.3 (centered by rs704017), which has been identified to be associated with colorectal cancer risk through GWAS. We plan to conduct fine-mapping of identified functional genes and search for novel colorectal cancer susceptible polymorphisms through case-control study. This functional analysis based fine-mapping strategy will lead to better understanding of GWAS-identified susceptible loci. Identified susceptible genetic variants could be used to colorectal cancer screening and will provide new clues for personalized prevention in the future.
