中文摘要: 高脂低蛋白饲料在水产养殖中应用已越来越广泛。在饲料中增加脂肪含量,可减少蛋白质用量,提高饲料利用率,减少蛋白氮向养殖水体的排放。但是,鱼类对脂肪需求和利用能力受生理条件的限制,饲料中过高脂肪会在鱼体内积累,脂肪转运发生障碍。本项目以团头鲂(Megalobrama amblycephala)为研究对象,通过高脂诱导,分析团头鲂肝脏组织载脂蛋白基因表达及DNA甲基化水平,探讨DNA甲基化对相关基因表达的调控作用,从表观遗传学角度出发,揭示团头鲂肝脏脂肪转运的表观遗传学机理。然后,通过在高脂饲料中添加胆碱,考察团头鲂载脂蛋白基因表达及DNA甲基化的变化水平,为通过营养干预手段优化团头鲂人工饲料配方提供科学依据。

英文摘要: Low-protein and high-lipid feed has now been widely adopted in the world aquaculture industry. An appropriate increase of dietary lipid could not only promote fish growth but also improve the feed utilization especially the protein utilization, thus exhibiting a protein-sparing effect and reducing nitrogenous losses to the environment. However, the optimal dietary lipid requirement of freshwater fish is relatively low. High dietary lipid might induce a dysfunction of lipid metabolism which consequently results in high body lipid deposition. It is reported that apolipoprotein plays an important role in lipid transport in fish. However, this aspect still remains poorly understood. Bearing this in mind, the present study was conducted to achieve the following aims (1) to investigate the gene expression and DNA methylation of apolipoprotein in the liver of Megalobrama amblycephala and (2) to evaluate the effects of choline on the methylation profile of apolipoprotein in Megalobrama amblycephala fed high dietary lipid. The results obtained here could partly characterized the lipid transport in Megalobrama amblycephala, and may also illustrate the mechanisms of fatty liver diseases observed in this fish and other herbivorous freshwater species at the molecular level.
