中文摘要: 阿尔茨海默症(AD)是一种严重影响老年人群日常生活的神经退行性疾病,发病率高,发病机理不明确,早期检测难度大,治疗成本高,给社会和家庭带来较大负担。本项目将应用新型纳米材料(生物分子功能化的金纳米粒子)为探针,借助纳米材料优越的光学性质,结合不同的光学(紫外可见光谱(UV-Vis)、共振光散射光谱(RLS)等)检测手段,发展简便、快捷、灵敏检测人体内淀粉样前体蛋白(APP)的方法,并对人体内APP含量与AD的相关性进行研究。探索和解决以下问题:(1)在实际样品中(如人体液,人脑脊液,神经细胞提取液等)高选择性、高灵敏度地对APP浓度进行定量分析;(2)对健康人和不同发病程度的AD患者体内的APP浓度进行统计学分析,研究AD的发病机理。上述研究有助于人们进一步认识以AD为代表的神经退行性疾病,为临床上AD的预防、检测和治疗提供帮助,对于以APP为靶点的新型药物的研制也具有重要意义。

英文摘要: Alzheimer's disease (AD) is a neurodegenerative disease, it serious disturb the daily life of old people. The incidence of AD is high, the pathogenesis is not clear, the early detection is difficult, and the treatment cost is high, all of these problems bring great burden on society and family. This project will use new nanomaterial (biomolecules functionalized gold nanoparticles) as probe, with the superior optical properties of nanomaterials, combined with different optical (UV-visible spectroscopy (UV-Vis), resonance light scattering spectroscopy (RLS), etc.) detection techniques, to develop a simple, fast, sensitive method to detect human amyloid precursor protein (APP), and to explore the linkage of APP expression level and AD development. We will focus on exploring and solving the following objects: (1) quantitatively determine the APP concentration in actual samples (e.g., human body fluid, human cerebrospinal fluid, nerve cell extract, etc.) with high selectivity and sensitivity; (2) using statistical analysis to study the different expression levels in healthy people and different AD patients, and to uncover the pathogenesis of AD.
