中文摘要: 武定鸡是云南省楚雄州独有的地区优势家禽品种,但其生长发育速度缓慢,难以推广养殖形成产业优势。因此,探究其生长发育的调节机制是一个重要课题。现已知,脊椎动物生长受神经内分泌生长轴上众多基因调节。本研究拟围绕家鸡生长轴上的两个关键因子:垂体生长激素(GH)及下丘脑来源的主要GH释放因子--生长激素释放激素(GHRH),采用多种分子生物学手段,初步探究: 1) GHRH受体在武定鸡垂体中的表达和细胞定位;2)武定鸡垂体GH mRNA丰度和血浆GH浓度在不同生长发育时期的变化规律;3) GHRH通过GHRH受体对武定鸡垂体分泌和合成GH的调控机理。研究结果可为探究GH和GHRH在楚雄武定鸡生长发育中的作用提供线索;同时,也为品种选育和挖掘地方优势种质资源奠定基础。

英文摘要: Wuding chicken is a local breed resource in Chuxiong, however, it is difficult to be raised in a large scale due to its slow growth rate. It is well documented that vertebrate growth is controlled by many genes in the hypothalamus-pituitary axis. Among them, pituitary growth hormone (GH) plays a pivotal role in controlling growth and development and its synthesis and release is regulated by multiple factors, particularly by hypothalamic growth hormone-releasing hormone (GHRH). To elucidate the roles of GH and GHRH genes in the growth and development of Wuding chickens, using multiple experimental approaches, present study aims to 1) investigate the pituitary cGH mRNA abundance and plasma GH concentration at different development stages of Wuding chickens; 2) localize GHRH receptors in Wuding chicken pituitary; 3) reveal how GHRH and its receptors are involved in controlling GH synthesis and secretion. Our study will not only shed lights on the role of GH and GHRH genes in controlling growth of Wuding chickens, but also be helpful for selection and breeding of local chicken strains.
