中文摘要: 红树林湿地是位于海陆过渡地带的特有生态系统,在防风消浪、维护热带亚热带海岸的生物多样性等方面发挥着重要的作用。近年来,由于人类生产和生活活动的加剧,红树林这一重要的生态系统正承载着来自陆源和海洋各种化学物的污染重负。红树林湿地微生物和环境污染物之间的相互作用是当今的研究热点。针对红树林湿地,国内外学者在多环芳烃、石油等污染物的微生物降解方面进行了大量的研究,然而,有关湿地微生物和抗生素污染之间相互作用的研究甚少。本项目拟综合运用新一代高通量测序技术、荧光定量PCR、多重PCR、基因组文库等多种分子生物学技术和微生物培养技术研究红树林沉积物细菌群落对典型β-内酰胺类抗生素污染的响应、寻找敏感响应指标;查筛未知的超广谱β-内酰胺酶基因、阐明其结构和功能。本项目研究成果将为红树林湿地污染生态学理论的完善积累关键数据,同时,将为解决β-内酰胺类抗生素正面临的严峻的细菌耐药性问题提供新的思路和途径。

英文摘要: Mangroves are important ecosystems located in tropical and subtropical regions. However, they are highly threatened ecosystems of anthropogenic influence, being exposed to many different pollutants. The interaction between mangrove microorganisms and environmental pollutants has become an important issue in recent years.Although numerous studies of microbial degradation of pollutants have been conducted in mangrove area, however, in those studies, researches were mainly focused on polynuclear aromatic hydrocarbons or oil. In the proposed project, we will investigate the changes of the mangrove bacterial community structure under antibiotics stress condition using high throughput sequencing technology.In addition,we will screen novel extended-spectrum β-lactamases genes using quantitative PCR, multiple PCR, cultivation methods and metagenomics technology. The present study will add useful informations to pollution ecology theory of mangroves and will provide useful novel ideas for treating bacterial resistance to β-lactam antibiotics.
