中文摘要: 鸭绿江滨海湿地处于我国海岸线最北端,湿地内形成了芦苇沼泽、碱蓬盐沼、潮沼、河口湾和浅海海域等复杂多样的生态系统,为东北亚最重要的鸟类迁徙停歇地,具有丰富的生物多样性,孕育着独特的放线菌资源。本项目首先根据北方滨海湿地的生态类型、潮汐规律及盐分梯度规律采集样品,建立湿地放线菌分离纯化方法,获得各种放线菌纯培养,分析可培养放线菌多样性及其生态分布特点。同时以各样品中的生物基因组总DNA为模板,采用PCR-DGGE和T-RFLPs技术,分析免培养放线菌多样性及其生态分布特点;最后,以农业生产上危害严重的瓜类枯萎病菌和烟草花叶病毒为靶目标,通过特定基因分析,结合菌碟法和叶碟法初筛,盆栽试验复筛,进行湿地放线菌活性评价;同时借助现代分离纯化和分析技术,进行农用活性化合物资源发掘。研究结果对揭示北方滨海湿地放线菌种群结构与环境因素间的关系,丰富我国放线菌资源并从中发掘新型农用活性化合物具有重要意义。

英文摘要: The littoral wetland along the Yalu River,located on the north end of the coastline in China, is the zone of a complex and diversified ecosystem which is composed of reed marsh, tidal marsh, seepweed marsh, estuarine marsh and so on.As the best-known resting place for migrators in northeast Asia, the coastal wetland has abundant biodiversity, and it is especially rich in special actinomyces.Firstly, the project will collect samples from the northen littoral wetland with consideration of the ecotype, tidal patterns and salt gradient, to establish the approach of actinomyces isolation and purification, which can lead to the culture of various actinomyces and the analysis of the diversity and distribution of cultivable ones.Secondly, with the adoption of PCR-DGGE and T-RFLPs, biological genome DNA of all the samples will be taken as model to do analysis of the diversity and distribution of the culture-free actinomyces.Lastly, targeting at the Melons fusarium oxysporium and tabacco mosaic virus seriously detrimental to agricultural production, the activity of the wetland actinobacteria will be evaluated through the analysis of the specific genes, after they are screened first by means of the cyclinder plate method and leaf disc method and then of the pot cultivation experiment.
