中文摘要: 高血压是一种最常见的心血管疾病,严重危害人类健康。然而,至今多数高血压患者的致病分子机理尚未阐明。Corin是一种跨膜丝氨酸蛋白酶,已经证实Corin的生物学功能为活化心钠肽(atrial natriuretic peptide, ANP),从而参与调节血压和水钠平衡。在前期研究中,我们已在中国人高血压性疾病患者中发现多个Corin基因变异。在功能研究试验中,这些基因变异使Corin活性下降。在本研究中,我们在高血压患者中筛选到了4个全新的Corin基因变异R349H、R530S、P866S和T924M,我们将通过细胞生物学及生化手段,阐明这些不同的基因变异对Corin蛋白生物合成、翻译后修饰及蛋白酶活性影响。我们的研究将有助于了解Corin蛋白质结构与功能的关系,从而进一步阐明Corin缺陷在高血压发病过程中的作用。

英文摘要: Hypertension is the most common cardiovascular disease. To date, however, the underlying molecular mechanism in most hypertensive patients remains unknown. Corin is a transmembrane serine protease, Our studies have established that the biological function of corin is to activate atrial natriuretic peptide (ANP), by which corin regulates blood pressure and salt-water balance. In our early studies, we have identified, for the first time, corin gene variations in hypertensive patients in China. In our functional experiments, these variations were shown to reduce corin biological activity. In this proposal, we have found 4 Corin gene variations R349H、R530S、P866S and T924M in hypertension patients, we plan to take cellular and biochemical approaches to elucidate how gene variations alter corin biosynthesis, post-translational modification, and protease activity. Our studies should help to understand corin protein structure and function and to define the role of corin defect in the pathogenesis of hypertensive disease.
