中文摘要: 牙周炎是成人失牙最主要的原因。在控制感染的基础上重建牙周组织是牙周病治疗的最终目标。然而迄今为止,牙骨质及其与牙周膜界面的修复再生还未能实现,牙周组织再生治疗技术尚不成熟。我们在前期研究中,采用静电纺丝技术制备了具有良好生物相容性的纳米羟基磷灰石涂层PCL/胶原纤维膜。同时获得一种新的重组牙骨质结构蛋白rhCEMP1,证实其具有诱导干细胞定向成牙骨质细胞分化、抑制向成骨细胞方向分化的作用。本项目拟在此基础上,采用三维直写电纺技术,制备具有牙周膜主纤维结构特征的PCL/胶原三维多孔膜,在其两面分别负载诱导成牙骨质细胞和成纤维细胞分化的细胞因子rhCEMP1和rhGDF5,形成具有双向诱导功能的三维多孔支架材料,复合牙周膜细胞膜片技术,实现工程化牙周组织的构建。本项目针对牙周炎患者牙周组织重建的问题,首次提出三维支架膜工程化牙周组织构建的新途径,为实现临床实用性牙周组织重构新技术打下基础。

英文摘要: Periodontitis is a major cause of tooth loss for adults. Periodontia reconstruction based on infection control is the key solution to periodontal treatment. However, the regeneration of cementum and periodontal ligament complex is still under research all over the world. In our previous study, we prepared a collage/PCL/nHA based fibers membrane with good biocompatibility by electrospinning. Moreover, a novel recombinant human cementum protein was produced and was proved to be able to induce stem cells differentiating towards cementoblast instead of osteoblast. Based on above work, the goal of the project mainly focuses on the fabrication of 3D porous fibrous film simulating the architecture of periodontal ligament based on the novel direct writing electrospinning. This 3D porous and biphasic guiding film is achieved by loading cytokine rh-CEMP1 on the one side and rh-GDF5 on the other side for inducing periodontal ligament cells to cementoblasts and periodontal fibroblasts correspondingly when periodontal ligament cell sheets are used to attaching to the scaffold. The project provides a novel approach to reconstruct periodontal tissue for periodontitis patients and will lay a solid foundation for periodontia reconstruction in clinical applicable.
