中文摘要: 菊科植物的头状花序造成了育种过程中的去雄障碍,从而限制了杂交育种的有效开展。雄性不育材料的获得将大大推进菊科植物新品种的培育进程。本课题旨在以花器官发生同源异型突变的万寿菊雄性不育两用系M525AB的花蕾为材料,进行转录组测序(RNA-seq)和表达谱测序(DGE),分离出与万寿菊花器官同源异型突变相关的差异ESTs,通过生物信息学分析和半定量RT-PCR技术,找到导致花器官同源异型突变的候选基因,结合RACE等方法获取候选基因的全长,利用realtime-PCR、RNA原位杂交技术和转基因技术完成候选基因在不育和可育万寿菊花序发育过程中的时空表达分析和功能验证。项目的实施不仅可以发掘与不育万寿菊花器官同源异型突变相关的基因,而且有助于揭示导致花器官同源异型突变的分子机理,为植物不育材料的创制提供借鉴。

英文摘要: TThe efficiency of cross-breeding was limited because of the labor costing procedures of emasculation of capitulum in the composite family. So, male sterile plants will greatly promote the breeding process of new varieties. In this project, taking the marigold male sterile two-type line M525AB, resulted from spontaneous floral organ homeotic conversion, as experiment material, the transcriptome sequencing (RNA-seq) and Digital Gene Expression Profiling (DGE) will be conducted to isolate difference ESTs related to the flower organ homeotic conversion. The candidate genes will be obtained through bioinformatics analysis and the semi-quantitative RT-PCR technology, and the full-length of the candidate genes will be acquired by the RACE method. Finally, the spatial and temporal expression pattern and gene function analysis of candidate genes will be carried out by Realtime-PCR, RNA in situ hybridization and genetic transformation during the inflorescence development process of sterile and fertile marigold. The implementation of the project could not only identifies the genes related to flower organ homeotic conversion in male sterile marigold, but also helps to reveal the molecular mechanism of homeotic conversion and to create the new male sterile plants.
